Also known as: S-Adenosyl-L-Methionine, Ademetionine
SAMe is a naturally occurring methyl donor synthesized from methionine and ATP. It is involved in over 100 methylation reactions and is clinically studied for mood support, joint health, and liver function.
S-Adenosylmethionine (SAMe) is a vital metabolite produced in every living cell, primarily in the liver. It serves as the principal methyl group donor in transmethylation reactions that regulate gene expression, neurotransmitter synthesis, phospholipid metabolism, and detoxification. SAMe donates its methyl group to become S-adenosylhomocysteine (SAH), which is subsequently converted to homocysteine and recycled via B-vitamin-dependent pathways. Clinical trials, particularly in Europe where SAMe has been prescribed for decades, demonstrate efficacy comparable to conventional antidepressants for mild-to-moderate depression and comparable to NSAIDs for osteoarthritis pain relief. SAMe also supports hepatic glutathione production and has been used adjunctively in liver disease.
SAMe is involved in the synthesis of serotonin, dopamine, and norepinephrine. Multiple randomised controlled trials show antidepressant efficacy comparable to tricyclics and SSRIs for mild-to-moderate depression.
SAMe stimulates proteoglycan synthesis in cartilage and has demonstrated analgesic and anti-inflammatory effects comparable to NSAIDs in osteoarthritis trials.
Split into 2 doses, taken on an empty stomach
Start with 200 mg twice daily and increase gradually. Full effects may take 2–6 weeks.
Split into 2–3 doses between meals
Effects may take several weeks to manifest; combine with B12 and folate for optimal methylation.
